摘  要:通过抑制率及抑制动力学的测定,研究了红旱莲总黄酮(HAF)对α-葡萄糖苷酶的抑制作用。利用对1,1-二苯基-2-三硝基苯肼(DPPH)自由基清除能力、β-胡萝卜素漂白抑制能力和抗DNA氧化损伤能力的测定,评估了红旱莲总黄酮体外抗氧化能力。红旱莲总黄酮是一种可逆的混合型α-葡萄糖苷酶抑制剂,其半抑制质量浓度(IC50)为0.237 mg/mL,抑制常数(K i)为0.149 mg/mL,酶–底物络合物抑制常数(K is)为0.123 mg/mL。红旱莲总黄酮有较强的DPPH自由基清除能力(IC50=2.71μg/mL),能够显著地抑制β-胡萝卜素的漂白及2,2'-偶氮二异丁基脒(AAPH)引起的DNA氧化损伤。红旱莲黄酮可作为潜在的抗氧化剂和预防治疗Ⅱ型糖尿病的药物或膳食功能因子进行深入研究与开发。The inhibitory effect on α-glucosidase of total flavonoids from Hypericum Ascyron L. (HAF) was investigated through the testing of inhibition rate and inhibition kinetics. The antioxidant activities of the flavonoids were evaluated in vitro by scavenging capability of DPPH free radical, the bleaching trials of beta-carotene, and the protecting of DNA oxidative damage. The result showed that the HAF was a reversible mixed inhibitor, and its half inhibition constant ICs0 was 0. 237 mg/mL, the inhibition constant Ki to a-glucosidase was 0. 149 mg/mL, and inhibition constant to the enzyme-substrate complex K~ was 0. 123 mg/mL, respectively. The antioxidant assay indicated that the HAF had significant DPPH radical scavenging activity (IC5o = 0. 27 ~g/mL), inhibition capacities on bleaching of G-carotene and DNA oxidative damage, which should be further investigated and utilized as a potential antioxidant and therapeutic drug of type I1 diabetes mellitus or dietary function factors.
