摘  要:为了选育出生产用的高产洛伐他汀红曲霉菌株,利用超高压对出发菌株进行诱变处理,并基于粗糙脉胞菌对洛伐他汀的生物敏感性,对菌株进行初筛,再通过HPLP法检测进行复筛。研究表明:超高压处理后,红曲霉总DNA出现了断裂和变性;选育出一株高产突变菌株HQ2,其在粗糙脉孢霉平板上抑菌圈直径达25mm,高出对照42.9%;产洛伐他汀量高达3.43mg/g,比对照组高52.4%;菌株的菌落颜色和形态也发生了较大变化。In order to select a strain with high-yield lovastatin production,the starting strains are mutagenized by ultra high pressure,which are primarily screened based on the biological sensitivity to lovastatin of Neurospora crassa,and then the secondary screening is conducted by HPLC.The results show that the total DNA of Monascus appears breakage and transgender after UHP treatment.At last,a strain HQ2 with the antibacterial circle diameter in Neurospora crassa plate as large as 25 mm is gained,which is 42.9% higher than that of the control group,the production level of lovastatin is 3.43 rmg/g,which is 52.4% higher than that of the control group.Meanwhile,the colony color and morphology of the strain change greatly.
