摘  要:乳酸菌是窖泥中的重要功能菌群,参与了浓香型白酒风味形成,因而探明窖泥中乳酸菌的种群特征,对于提高白酒品质具有重要意义.采用PCR-DGGE技术对泸州老窖不同窖龄窖泥乳酸菌种群进行了分析.结果显示,根据DGGE图谱条带情况,不同窖泥样品中乳酸菌多样性差异明显;多样性指数分析结果表明,样品的Patrick丰富度指数R介于14-19,Shannon-Wiener指数H值介于2.634 5-2.888 9之间,Simpson指数C值介于0.055 7-0.071 9.根据图谱选取16条DGGE条带进行序列分析,它们分属于Lactobacillus(乳杆菌属)、Pediococcus(片球菌属)、Weissella(魏斯氏菌属)、Enterococcus(肠球菌属)、Exiguobacterium(微小杆菌属)、Bacillus(杆菌属)、Rummeliibacillus属以及Cohnella属,其中Lactobacillus(乳杆菌属)为优势菌属,L.acetotolerans(耐酸乳杆菌)为窖泥中乳酸菌的优势种群.Lactobacillus is an important functional bacterium in pit mud, participating in the formation of strong aromatic liquor. Therefore exploring lactic acid bacteria species in pit mud is of great significance for the improvement of liquor quality. In this study, seven cellar pit mud samples were collected from different-aged cellars in Luzhou Laojiao Liquor Co. Ltd. The DNA was isolated, and the diversity of lactic acid bacteria was analyzed by PCR-DGGE technique. The results of DGGE showed significant diversity of lactic acid bacteria in the pit muds. Analysis of the diversity index showed the Patrick richness index R between 14 and 19; Shannon-Wiener index H varied between 2.634 5 and 2.888 9; and Simpson index C was between 0.055 7 and 0.055 7. Furthermore, 16 representative DGGE bands were picked, rinsed, and then amplified by using primers LAC1/LAC2, the PCR fragments were cloned and sequenced. The results revealed that the lactic acid bacteria in different-aged cellar pit mud samples were very abundant, belonging to genus Lactobacillus, Pediococcus, Weissella, Enterococcus, Exiguobacterium, Bacillus, Rummeliibacillus, and Cohnella, respectively. The predominant lactic acid bacteria among all the pit muds were Lactobacillus acetotolerans.
