摘 要:运用水酶法提取火麻仁油,依次选用Alcalase2.4L(碱性蛋白酶)、Protamex(复合蛋白酶)、Neutrase0.8L(中性蛋白酶)、Celluclast1.5L(复合纤维素酶)、Viscozyme(复合植物水解酶)单独并复配对云麻一号和巴马火麻仁酶解提油,以提油率为指标,最终选取Alcalase2.4L提取云麻一号。研究了粉碎次数、液料比、酶用量、酶解初始pH、酶解温度和酶解时间对提油率的影响,并用响应面法进行了优化。得到的最优酶解条件为:粉碎次数4次(5 s/次),液料比3.5∶1,酶用量2.0%(以火麻仁质量计),酶解初始pH 7,酶解温度70℃,酶解时间3.5 h。在最优条件下,火麻仁的提油率可达83.81%。The aqueous enzymatic extractions oi heepseed oils trom Yunma No. 1 and Bama Huoma were researched with Alcalase2.4L, Protamex, Neutrase0.8L, Celluclastl. 5L and Viscozyme respectively or compositively. According to the oil extraction rate, Alcalase2.4L was selected as the best enzyme to hydrolyze Yunma No. 1. Then, the effects of crushing times, ratio of water to material, enzyme dosage,initial enzymolysis pH, enzymolysis temperature and enzymolysis time on the oil extraction rate of hempseed were studied. The optimal enzymolysis conditions were obtained by response surface methodology as follows: crushing times 4(5 s for each times) ,ratio of water to material 3.5:1 ,enzyme dosage 2.0% (based on the mass of heepseed), initial enzymolysis pH 7, enzymolysis temperature 70 ~C and enzymolysis time 3.5 h. Under these conditions,the oil extraction rate of heepseed was 83.81%.
