摘 要:建立乳制品中五种核苷酸的含量测定方法。通过对实验前处理条件、色谱柱、流动相等条件的选择,确定了乳制品中核苷酸的检测分离条件为:SUPELCOSIL LC-18-T色谱柱;检测波长:254 nm、278nm;流动相为含0.75 mL四丁基氢氧化铵溶液的磷酸二氢钾水溶液(pH 4.5)和甲醇。实验表明:五种核苷酸在1~20μg/mL的范围内均呈线性,且相关系数均在0.9999以上;三个水平的加标回收率均在98~107%之间,且RSD均小于4%。Establish a method of determination for nucleotides contents in milk products. Based on the tests in different chromatographic column and mobile phase, the best detect conditions for nucleotides in milk products were confirmed. SUPELCOSIL LC -18 -T column was used for isolation and the nucleotides were determined quantitatively at UV254 nm and 278 nm wavelength with HPLC. The mobile phase was phosphate (with Tetrabutylammonium hydroxide solution, pH = 4. 5) and methanol. The five nucleotides were all linear in the concentration range of 1 - 20 μ/mL ( R2=0.9999). The average recovery rate (n = 6) were between 98% -107 % , RSD 〈 4 %.
