摘  要:采用正交试验优化木糖葡萄球菌A2的增殖培养基配方,并确定其高密度培养条件和高密度培养物的富集条件。通过单因素试验确定木糖葡萄球菌A2液体培养基的最佳碳源为乳糖,最佳氮源为示蛋白胨,最佳营养因子为酵母膏和香菇汁;利用L9(34)正交试验得出增殖培养基的最佳配方为0.5%乳糖、2%示蛋白胨、0.2%酵母膏、5%香菇汁和7.5%NaCl。木糖葡萄球菌A2高密度培养的最适初始pH值为7.5,最适装液量为90%。以最佳培养基和培养条件在32℃培养17 h后,菌液中活菌数可达到3.43×109CFU/mL。对高密度培养物进行富集的最佳条件为6 000 r/min离心30 min,离心后菌种存活率为82.5%,浓缩倍数为74.48倍,活菌菌密度可达2.07×1011CFU/g。本研究成果可为生产发酵肉制品的直投式发酵剂提供优良菌种。Orthogonal experiment was conducted to optimize the enrichment media formula of Staphylococcus xylosus.The high-density culture conditions of Staphylococcus xylosus and the concentration parameters of high-density cultures were also determined. The single factor experiment results showed that the optimal carbon source and nitrogen source were lactose and proteose peptone, respectively; and the optimal nutritional factor were yeast extract paste and mushroom juice. The L9(34) orthogonal experiments results showed that the optimal formula of the enrichment media was consist of0.5% lactose, 2% proteose peptone, 0.2% yeast extract paste, 5% mushroom juice and 7.5% NaCl. The optimal initial pH for the high-density culture of Staphylococcus xylosus A2 was 7.5 and the optimal liquid volume in flask was 90%.Under the optimal conditions, the maximum viable counts of the strain reached 3.43×109CFU/mL after cultured at 37 ℃for 12 h. After centrifugalized at 4 000 r/min for 30 min, the maximum starter survival rate of 82.5% was observed, and there were 74.48-fold of enrichment. The finally viable count was 2.07 ×1011CFU/g. The present research could supply ready-to-use starter for the production of fermented meat products.
