摘 要:采用外翻肠囊法和大鼠在体单向灌注法,以肠吸收速率(Ka)和表观渗透系数(Papp)为指标,考察豆豉纤溶酶纳米脂质体的小肠吸收部位及效率。结果显示,由外翻肠囊法测得豆豉纤溶酶纳米脂质体的回肠段Ka和Papp分别为3.25×10-2μg/s和2.355×10-6cm/s,且吸收速率随着浓度的升高而降低,最终出现饱饮现象。由大鼠在体单向灌注法测得的Ka和Papp分别为(1.49±0.22×10-3)μg/s和(7.90±1.46)×10-5cm/s。上述两种方法均表现出豆豉纤溶酶纳米脂质体在回肠的吸收效率为最高,这可能与回肠中大量存在Peyer’s淋巴结有关,而Peyer’s淋巴结中存在的M细胞可能导致饱饮现象。Using everted gut sac method and the single-pass intestinal perfusion in rats method, and the absorption rate and apparent permeability coefficient of indicators, we examined the site and efficiency of intestinal absorption of douchi fibrinolytic enzyme nanoliposomes. The results showed that: Both methods are measured uptake of DFE nanoliposomes largest in the ileum. Measured by everted sacs absorption rate (Ka) and the apparent permeability coefficient (Papp) were respectively 0.0325 μg/s and 2.355×10^-6cm/s, and the absorption rate of DFE nanoliposomes decreased with increasing concentration, the phenomenon occurred full drink; Ka and Papp were respectively (1.49±0.22×10^-3) μg/s and (7.90±1.46)×10^-5 cm/s by the single-pass intestinal perfusion in rats method. The absorption of DFE loaded nanoliposomes were better in the ileum than in the other intestinal segments by above two ways. These may be related to the lots of Peyer's patchs exsisted in ileum. The M ceils in Peyer's patchs lymph node might cause the full drink phenomenon along with the increase of the concentration.
