摘 要:通过酶法与膜分离技术和沉淀技术的有效结合,改善肝素钠提取工艺,获得高效价的肝素钠精品。以猪小肠黏膜作为原料,通过酶解-树脂吸附/洗脱-膜分离-乙醇沉淀-干燥的生产工艺方法,从中提取肝素钠。与传统的盐解-离子交换-乙醇沉淀法相比,酶法与膜分离技术和沉淀技术的有效结合,可以使分离获得的肝素钠效价提高至160 U/mg,总效价比传统方法提高近64%。酶法与膜分离技术和沉淀技术相结合,有助于提高肝素钠分离的收率和效率,降低生产成本,工艺合理可行。The effective combination of enzymatic, membrane separation and precipitation technology was applied to improve the extraction technology of heparin sodium and obtain heparin sodium products with higher titer. Heparin sodium was extracted through enzymatic hydrolysis-resin adsorption/des-membrane separation-ethanol precipitation-production protocol from pig small intestinal mucosa. Compared to traditional protocol of salt solution-ion exchange-ethanol precipitation, the combination of enzymatic and membrane separation, improved the separation of heparin potency to 160 U/mg and nearly 64% higher than traditional method. The method of combination of enzymatic, membrane separation and precipitation technology helped to increase the efficiency and titer of heparin sodium and reduce production cost.
