摘  要:为优化青霉1407产纤维素酶发酵培养基,采用单因素试验法寻找青霉产纤维素酶所需培养基的碳源氮源、有机氮与无机氮的比例、诱导物浓度以及金属离子种类对产酶的影响,然后利用响应面分析法确定关键影响因子的最佳参数,结果显示:在麸皮2.955%、药媒3.25%、微晶纤维素3.75%、硫酸铵1.50%、硫酸镁0.03%、氯化钙0.03%条件下,青霉产纤维素酶的理论酶活最大值达到6 968.02 U/mL。验证试验纤维素酶活为(6 954.32±5.23)U/mL,与预测值接近,说明建立的回归方程模型与验证试验结果相符。The impact factor of the culture medium have been optimized for producing cellulose by Penicillium sp. 1047. Firstly through the monofactorial experiments, the kind and concentration of carbon source and the kind of nitrogenous source, the proportion of organic nitrogen and inorganic nitrogen, the concentration of inducer and the kind of metal ion for the influence were studied for producing cellulose, then the optimized parameters of main factors were deter mined by response surface methodology. The optimal culture medium formula (W/V): bran 2.955%, medicine medium 3.25%, microcrystalline cellulose 3.75%, ammonium sulfate 1.5%, calcium chloride 0.03%, the theoretical value of cel- lulose activity were 6 968.02 U/mL. Repeated experiment were done and the cellulose activity were 7 109.22 U/mL which had closed to the predicted value, and the regression equation model consistent with the experimental results.
