摘  要:分析了小鼠感染大肠杆菌O157:H7及补充副干酪乳杆菌KLDS1.0351和植物乳杆菌KLDS1.0986后小鼠肠粘膜中细胞因子IL-2、IL-6、IFN-γ及s Ig A的变化,结合小鼠的表观特征探究两株乳酸杆菌对大肠杆菌引起的疾病的治疗作用。将小鼠分为空白组、对照组、KLDS1.0351和KLDS1.0986,建立大肠杆菌模型成功后,再连续补充两株乳酸杆菌一周,采集小肠用ELISA试剂盒测定上述3种细胞因子及s Ig A的质量浓度。结果表明:KLDS1.0351和KLDS1.0986能够提高感染大肠杆菌小鼠的体质量;感染大肠杆菌后,s Ig A和IL-6在第5天达到最大值,第7天开始下降,IFN-γ和IL-2在第3天达到最大值,在第5天开始下降。补充两株乳酸杆菌后,3种细胞因子和s Ig A的质量浓度升高,且与其他两组差异显著。本试验证实KLDS1.0351和KLDS1.0986能对大肠杆菌感染引起的腹泻和体质量减轻起到缓解的作用,两株菌株能显著促进感染大肠杆菌小鼠肠粘膜中IFN-γ、IL-2、IL-6及肠道中s Ig A的分泌,提高机体免疫力。The possible immunomodulatory effect of Lactobacillus paracasei KLDS1.0351 and Lactobacillus plantarum KLDS1.0986 were evaluated on the intestinal mucosal immunity in the mice infected with Escherichia coli. Trial was divided into four treatments, challenged treatment with Escherichia coli for seven days, then infected animals treated with Lactobacillus paracasei and Lactobacillus plantarum for another one week. SIg A and cytokines(IFN –γ,IL-2,IL-6) were determined by ELISA. Lactobacillus paracasei KLDS1.0351 and Lactobacillus plantarum KLDS1.0986 attenuated the influence of Escherichia coli by the high weight gain. After treatment with Escherichia coli, the levels of IL-6 and s Ig A increased and reached to the maximum value at the fifth day, decreased at the seventh day. IFN-γand IL-2 increased and reached to the maximum value at the third day, decreased at the fifth day. The levels of assayed cytokines and s Ig A were significantly increased after supplementing Lactobacillus paracasei KLDS1.0351 and Lactobacillus plantarum KLDS1.0986 at eighth day, then remained higher level than pathogenic and control groups. Lactobacillus paracasei KLDS1.0351 and Lactobacillus plantarum KLDS1.0986 enhanced protection against Escherichia coli infection, and improved the intestinal mucosal by stimulating s Ig A and cytokines production.
