摘  要:为了获得能用于茶皂素组分分析及鉴定的高纯度茶皂素,采用有机溶剂沉淀法、乙醇重结晶法、大孔树脂法和反相柱层析法4种方法对冷榨油茶籽饼中的茶皂素进行纯化,以茶皂素紫外吸收特征峰强度为指标,比较4种方法的纯化效果。结果表明:反相柱层析法能有效去除茶皂素粗品中大部分杂质,是获得高纯度茶皂素的有效方法,得到的茶皂素样品纯度为95%,可作为茶皂素的标准品。利用高效液相色谱和质谱对茶皂素反相柱层析纯化物进行组分分析,验证了其中5种组分为文献中已经报道的茶皂素单体。The organic solvent precipitation method, ethanol recrystallization method, macroporous resin method and reversed phase column chromatography method were used to purify tea saponin prepared from cold pressed oil - tea camellia seed cake so as to obtain high purity tea saponin for the analysis and iden- tification of tea saponin components. The purification effects of the four methods were compared with vio- let characteristic absorption band intensity as indicator. The results showed that the reversed phase col- umn chromatography method was an effective way to remove most of the impurities and obtained high puri- ty tea saponin. The purity of prepared tea saponin sample reached 95 % , which could be used as tea sap- onin standard. The component analysis of tea saponin sample obtained by reversed phase column chroma- tography method by HPLC and MS indicated that the structures of five components were the same as those in previous report of tea saponin monomers.
