摘 要:采用单频逆流超声技术对东北黑蜂蜂胶黄酮进行提取,研究了乙醇体积分数、温度、提取时间、液料比和占空比对黑蜂蜂胶有效成分总提取率和蜂胶中黄酮提取率的影响。在单因素试验的基础上,采用Box-Behnken中心组合试验设计对逆流脉冲超声辅助提取条件进行优化,得到最佳提取参数为:乙醇体积分数80%、提取温度51℃、提取时间16 min、液料比30︰1(mL/g)、占空比10 s/5 s,在此条件下黄酮提取率为38.72%。测定相同条件下超声提取液与未超声提液的抑菌活性,超声的蜂胶提取液和未超声的蜂胶提取液对金黄色葡萄球菌、李斯特菌、枯草芽孢杆菌均有抑菌性。The counter-current pulsed ultrasound technology was adopted to extract the propolis' flavonoids from the northeast China. Study the influence of the ethanol concentration, temperature, extraction time, solid-liquid ratio and duty ratio to the propolis' flavonoids. The Box-Behnken central composite design combined with response surface analysis was used to optimize the pulsed ultrasound-assisted extraction condition. In the study, the regression equation was got and proved through regression analysis and experimental verification. The results showed that the optimum conditions of counter-current pulsed ultrasound-assisted extraction were ethanol concentration of 80%, temperature of 51 ℃, time of 16 min, solid-liquid ratio of 30︰1(mL/g) and duty ratio of 10 s/5 s. The total yield of propolis flavonoids from the Northeast China could reach up to 38.72%. In the same conditions, measure the extracts' antimicrobial activities which were extracted by counter-current pulsed ultrasound technology and the traditional extraction technology. The extracts had antimicrobial activities on Staphylococcus aureus, Listeria monocytogenes and Bacillus subtilis.
