摘  要:研究了胶原蛋白肽氨基酸组成对其在小鼠体内吸收程度的影响。利用离子交换层析分别制备出富含碱性和酸性氨基酸的胶原蛋白肽,小鼠灌胃后检测了血清中羟脯氨酸和胶原蛋白降解后的多肽相对丰度的差异。氨基酸组成分析结果表明阳离子交换层析制备的胶原蛋白肽(CPC)富含精氨酸(Arg)和赖氨酸(Lys),含量比未经过层析分离的多肽(CPo)分别高12.2%和7.4%。阴离子交换制备的多肽(CPA)中Arg和Lys含量比CPo低18.3%和26.5%。通过离子阱质谱技术从CPC中检测到9种含Arg或Lys的多肽,其含量远大于CPA中的含量。动物实验结果表明CPC在小鼠体内吸收高于CPA,小鼠血清中CPC在吸收峰值时Hyp的浓度是CPA的1.39倍,采用三重四级杆质谱(TSQ)从CPC灌胃后小鼠血清中检测出含Hyp的寡肽(Ser-Hyp-Gly、Pro-Hyp-Gly、Leu-Hyp),3种寡肽分别是CPA灌胃后小鼠血清中浓度的2.1、1.48和1.51倍,初步证明了氨基酸组成是影响胶原蛋白肽在小鼠体内吸收的重要因素。Effect of amino acid composition in collagen peptides on the intestinal adsorption in mice was investigated. Collagen peptides( CPO),with mean MW 1 k Da,were treated using cation and anion exchange chromatography,and the eluted peptides were named as CPCand CPA,respectively. The amino acid compositions in CPCand CPAwere determined. The intestinal absorption of peptides by mice was determined by detecting plasma hydroxyproline( Hyp) and di / tri-peptide after intragastric infusion. The result indicated the Arg and Lys contents in CPCwere 12. 2% and 7. 4% higher than those in CPO,and the Arg and Lys contents in CPAwere18. 3% and 26. 5% less than those in CP" C. More Arg / Lys-containing peptides were found in CPCthan that in CPAby HPLC-MS analysis. The result of animal test indicated that the Hyp content in mice plasma after intragastric infusion of CPCwas 1. 39 times higher than that of CPA. Oligopeptides,Ser-Hyp-Gly,Pro-Hyp-Gly and Leu-Hyp,were found in mice plasma. The plasma concentrations of three oligopeptides were 2. 1,1. 48 and 1. 51 higher after infusion of CPCcompared to CPA. This research suggests that the amino acid composition is a key factor affecting peptide absorption in intestine.
