毕赤酵母产木聚糖酶发酵条件优化及其酶学特性研究

摘  要:本研究在50L发酵罐水平上对毕赤酵母产木聚糖酶发酵条件进行优化,并对其酶学特性进行研究。利用单因素试验系统考察了微生物种龄、诱导时机、诱导培养温度和诱导时期pH值对产木聚糖酶的影响。结果表明:在最佳种龄23h、发酵液细胞湿重121g/L左右开始诱导,诱导培养温度26℃,诱导时期维持pH值5.6的优化工艺条件下,木聚糖酶酶活可达到47570U/mL;酶的最适反应pH值为5.0~6.0,且具有较好的耐热性,85℃作用2min后酶活收率仍可达到87.8%,能够用于饲料高温制粒。The optimization of fermentation conditions and the enzymatic properties of xylanase produced by Pichia pastoris were studied in 50 L fermentor.The effects of cell age,the time of induction,temperature and pH of induction culture on expression of xylanase were investigated.The optimum parameters of xylanase fermentation were:the optimal cell age of 23 h, methanol added when wet cell weight reached 121 g/L,induction temperature of 26℃,maintain pH of induction culture 5.6.Under above conditions,the xylanase reached an activity of 47570 U/mL.The optimal working pH value of xylanase was 5.0 - 6.0,and it had good heat resistance,the rate of enzyme activity could remain 87.8% at 85℃ for 2 rain. The xylanase could be used for feed granulation at high temperature.
