摘 要:建立超高效液相色谱法(UPLC)、二极管阵列检测器(PAD)快速测定葡萄酒中防腐剂脱氢乙酸(DHA)的方法。采用甲醇-0.02 mol/L乙酸铵溶液(体积比=20∶80,p H=7.5~8.5)作为流动相,流速为0.25 mL/min,柱温为35℃,检测波长为293 nm。该方法以保留时间(Rt)定性,外标法定量,在5.0 mg/L^200.0 mg/L范围内待测组分的峰面积与浓度有良好的线性关系,R2>0.999 9,样品在5.0、10.0、25.0 mg/L 3个加标水平回收率范围为87.0%~101.2%(n=10),相对标准偏差小于3%,该方法的定量限(LOQ)为5.0 mg/L。A method was developed for the rapid determination of dehydroacetic acid(DHA) in wine. The sample was tested by ultra performance liquid chromatography(UPLC) with diode array detector(PDA). The mobile phase was methanol-0.02 mol/L Ammonium acetate(mL∶mL=20 ∶ 80,pH=7.5-8.5),flow rate was 0.25 mL/min,column temperature was 35 ℃,determine wavelength was 293 nm. The method for qualitative analysis was based on retention time. And external standard method was used for calculation. Good linear relationship was achieved in the range of 5.0 mg/L-200.0 mg/L for DHA with correlation coefficients above 0.999 9. The recoveries of the method ranged from 87.0 %-101.2 %(n=10) with the relative standard deviations(RSDs) less than 3 % at3 spiked levels of 5.0, 10.0, 25.0 mg/L. The limit of quantification(LOQ) was 5.0 mg/L for DHA.
