摘  要:目的建立一种快速测定茶鲜叶中β-胡萝卜素的高效液相色谱(HPLC)方法。方法以丙酮为提取溶剂;色谱柱为SunFire^(TM)C_(18)(150 mm×4.6 mm,5μm);以三氯甲烷-乙腈溶液(9:1,V:V)为流动相,等梯度洗脱;波长450 nm;流速1.0 mL/min;柱温35℃。结果在该色谱条件下β-胡萝卜素与其他物质得到了有效的分离,且β-胡萝卜素在9.5×10^(-3)~0.76μg范围内与峰面积有良好的线性关系(r=0.99997),回收率97.32%~101.26%。结论该方法重复性好、精密度高、回收率较好,可作为茶鲜叶中β-胡萝卜素的定量测定方法。Objective To develop a determination method for β-Carotene in fresh tea by high performance liquid chromatography(HPLC).Method The β-Carotene was extracted from fresh tea leaves by acetone.A SunFire^(?) C_(18) column(150 mm×4.6 mm,5 μm) was used with the mobile phase of chloroform-acetonitrile solution(9:1,V:V).The flow rate was 1.0 mL/min,wavelength was 450 nm and the column temperature was35 ℃.Result This method showed a good linearity(r=0.99997) for β-Carotene(ranging from 9.5×10^(-3) to0.76 μg) and the recovery of β-Carotene was from 97.32%to 101.26%.Conclusion This method is precise,accurate and suitable for the determination of β-Carotene in fresh tea leaves.
