摘  要:以木醋杆菌(Acetobacterxylinum)为发酵菌种,通过Plackett-Burman试验设计确定了陈米糖化液培养基中酵母膏、KH2PO4、Fe SO4、乙醇对木醋杆菌发酵产细菌纤维素具有显著影响,并采用Box-Behnken试验设计对各显著影响因子进行优化,获得最优的陈米糖化液发酵培养基配方为:在陈米糖化液培养基基料中加入酵母膏13.1 g/L、蛋白胨10 g/L、KH2PO45.7 g/L、Mg SO43.1 g/L、Fe SO40.3 g/L、柠檬酸0.3 g/L、无水乙醇4.0%。在此优化条件下,细菌纤维素的产量为7.08 g/L,是陈米糖化液培养基基料发酵产细菌纤维素(0.38 g/L)的18.6倍,比基础发酵培养基细菌纤维素产量(4.80 g/L)提高了47.5%。Using Acetobacter xylinum as fermentative strain, yeast extract, KH2PO4, FeSO4 and ethanol had obvious effect on bacterial cellulose pro- duced fi'om A. xylinum by Plackett-Burman experiments. The factors were optimized by Box-Behnken experiments, and the optimum formula of stocked rice saccharification liquid fermentation medium were yeast extract 13.1 g/L, peptone 10 g/L, KH2PO4 5.7 g/L, MgSO4 3.1 g/L, FeSO4 0.3 g/L, citric acid 0.3 g/L, absolute ethanol 4.0%. Under the conditions, the yield of bacterial cellulose was 7.08 g/L, which was 18.6 times of the bacterial cellulose yield (0.38 g/L) in the basic material of stocked rice saccharification liquid medium and was increased by 47.5% than that (4.80 g/L) of basis fermentation medium.
