摘  要:为了脱除中性蛋白酶发酵液中的色素,降低色素在产品应用中的影响,研究并比较了树脂、活性炭、H2O2对中性蛋白酶发酵液的脱色效果。实验结果表明,阴离子交换树脂D380(Cl)型能使其有效脱色并保留较高的蛋白酶活性;利用响应面实验建立数学模型,并确定D380(Cl)型树脂脱色最适流速为2.35柱体积/h(bed volume/h,BV/h),最佳上样量为0.4 BV,最适上样酶活力为40 000 U/m L,最适p H值为6,此时,中性蛋白酶发酵液脱色率为86%,酶活回收率为95%。D380(Cl)型树脂连续使用10次仍有较强脱色能力,脱色率在80%以上,酶活回收率在90%以上。To remove the pigments in neutral protease fermented broth, a simple method for the decoloration of neutral protease fermented broth was developed through different medium. Iron exchange resin D380 (C1) demonstrated the highest decoloration efficiency and good protease recovery. Under the optimized dynamic adsorption conditions through single factor method and response surface methodology ( pH 6, uploading and elution flow rate of 2.35 BV/h, 0.4 BV of 40 000 U/mL enzyme solution), the ratios of decoloration and protease yield for D380 (C1) were 86% and 95% , respectively. Used D380 (C1) could be regenerated easily, and the regenerated D380(C1) was as effective as not used one. After 10 times reuses of regenerated D380(C1) , the ratios of decoloration and protease yield could also reach 80% and 90% , respectively. It provides a potential approach for large-scale production of neutral protease.
