摘 要:对热水浸提法提取的海湾扇贝多糖采用乙醇沉淀法、TCA法、乙醇沉淀法+TCA法等方法脱除海湾扇贝多糖中的蛋白质并纯化,结果表明,乙醇沉淀法+TCA法有很好的脱蛋白效果,累计蛋白质脱除率达到91.8%,而多糖损失率只有10.96%。脱蛋白的海湾扇贝多糖经DEAE-52纤维素柱层析分离得到两种组分,其中酸性多糖为主要成分。利用纯化分离的海湾扇贝多糖对体外培养的人卵巢上皮癌细胞株肿瘤细胞的抗肿瘤作用进行评价。经20 mg/m L海湾扇贝多糖处理,SKOV3肿瘤细胞的微观结构和超微结构均发生改变,表现为细胞皱缩,透明度降低,细胞密度降低,膜结构改变,核结构改变,核内染色质固缩等,并可对细胞周期产生不同程度的阻滞作用。海湾扇贝多糖具有一定的体外抗肿瘤活性。In the present study, ethanol precipitation, trichloroacetic acid(TCA) and their combination were used to remove the protein from scallop polysaccharide extracted by hot water. The results showed that the combination of ethanol precipitation and TCA was the most effective method to remove protein from scallop polysaccharide in which the accumulated percentage of deproteinization was 91.8% while the loss of polysaccharide was 10.96%. Moreover, the results obtained by DEAE-52 cellulose ion exchange chromatography showed that acidic components were the major constituents of bay scallop polysaccharide. In addition, the antitumor activity against SKOV3 cells of scallop polysaccharide was evaluated in vitro. After the treatment of bay scallop polysaccharide of 20 mg/m L, the microstructure and ultrastructure of SKOV3 cells were changed, characterized by cell shrinkage, transparency decrease, membrane structure change, nuclear structure change and nuclear chromatin pyknosis, and the cell cycle was blocked in varying degrees. The results suggested that polysaccharide from bay scallop had certain antitumor activity in vitro.
