拟青霉发酵生产胞外多糖的研究

摘  要:探讨拟青霉属产胞外多糖和菌丝体生物量最大时的液体培养条件的优化,主要考察培养基组分、碳氮比和物理参数这三方面的影响。结果表明,最适培养基配方为葡萄糖30 g/L、酵母膏20 g/L、KH_2PO_40.5 g/L,Mg SO_40.1 g/L。最适培养条件为27℃,摇床转速400 r/min,培养时间为7 d。在此条件下,拟青霉属胞外多糖的产量为2.51 g/L,菌丝体生物量为23.5 g/L。The liquid culture conditions were optimized for maximal production of mycelial biomass and exo- polysaecharide by Paecilorayces japonica. The effects of medium composition, C/N ratio and physical parameters were investigated. From these experiments, 30 g/L glucose, 20 g/L yeast extract, 0.5 g/L KH2PO4, and 0.1 g/L MgSO4 in distilled water were found to be the most suitable carbon, nitrogen, and mineral sources, respectively. The optimal temperature, initial pH, agitation, and aeration were determined to be 27 ℃, uncontrolled pH, 400 r/rain and for 7d, respectively. Under these optimal conditions, the maximum mycelial growth and polysaceharides production were 2.51 g/L and 23.5 g/L, respectively.
