摘 要:建立了测定黄骨鱼皮肤中3种合成着色剂(柠檬黄、日落黄和喹啉黄)和3种天然着色剂(叶黄素、β-胡萝卜素和虾青素)的分析方法。样品采用超声波细胞破碎法进行提取,提取不同的着色剂选用不同的提取剂。以叶黄素含量为指标,通过单因素试验确定了超声波细胞破碎法的最佳提取条件:全程时间8 min,超声/间隙时间2s/3s,超声功率比55%,保护温度35℃。采用C18柱(4.6 mm×250 mm,5μm),流速为1.0 m L/min,检测波长为446nm,分离不同的着色剂选用不同的流动相。合成着色剂和天然着色剂分别在2.5~50 mg/L和1.6~32 mg/L范围内呈良好线性,相关系数均大于0.9995。回收率在95.8%~102.5%之间,方法检出限在0.3~1.0 mg/kg之间。用该方法测定了12批次黄骨鱼,均检出含有叶黄素,其余5种未检出。A new method for the determination of three kinds of artificial pigments ( tartrazine, sunset yellow and quinoline yellow) and three kinds of natural pigments (lutein, 13-carotene and astaxanthin ) in the skin of Pelteobagrus fulvidraco was firstly proposed. Samples were extracted by ultrasonic ceil-break method. Different extraction solvents were selected according to different pigments. Based on the content of lutein, the experimental conditions were optimized through the single-factor experiment: full work time 8 min, work/terral time 2s/3s, ultrasonic power ratio 55% and temperature 35~C. HPLC separation was conducted on a C1s column (4. 6 mm ~ 250 mm, 5 i^m) at a flow rate of 1.0 mL/min and the detection was carried out at a wavelength of 446 nm. Different mobile phases were selected according to the separation of different pigments. The linear ranges of the method for artificial pigments and natural pigments were 2. 5 - 50 mg/L and 1.6 - 32 mg/L respectively with correlation coefficients not less than 0. 9995. The recoveries of the pigments were between 95.8% and 102. 5% , and the detection limits of the pigments for this method were between 0. 30 and 1.0 mg/kg. The method was applied in the determination of 12 batches of Peheobagrus fulvidraco. The lutein was detected and none of the rest of five pigments was detected. This method is found to be convenient, efficient and accurate, and could meet the determination of related pigments in the skin of Peheobagrus fulvidraco.
