摘  要:目的探讨新生儿疾病筛查滤纸干血片长期保存后TSH和Phe的稳定性。方法对2007-2011年间4℃保存的500份新生儿足跟血滤纸干血片,分别用时间分辨荧光免疫分析法和荧光计数分析法,常规检测和延长洗脱时间检测TSH和Phe水平,与原始检测结果进行对比分析。结果 4℃保存血片的Phe浓度值,与2007-2011年间的原始测定值比较,差异无统计学意义,P>0.05;TSH常规方法检测新值与原始测定值比较,保存1-2年组,差异无统计学意义,P>0.05,保存3-5年组,差异有统计学意义,P<0.01,延时法重复检测3-5年组血片,检测值与原始值比较,其差异有统计学意义,P<0.01。结论4℃保存1-5年血片Phe检测值与保存时间、洗脱时间无明显关系,与原始值无明显差异;血片保存两年内,TSH检测值可以保持稳定,保存3年及以上,检测值与原始值差异显著,且延长洗脱时间不能使TSH检测值恢复至原始水平。Objective:To discuss the stability of TSH and Phe detected value of neonatal dried blood filter cards after longterm storage and the accuracy of the original sample review. Methods:We use 500 newborn dried blood filter cards stored in 4℃from 2007 to 2011.Detected the TSH and Phe concentration levels of all the dried blood spots(DBS),with time-resoveled fluorescence immunoassay analysis method and fluorescence counting method respectively,conventional detected and prolong the elution time detected the DBS kept under different environment,compared with the original values,and analysis. Results:The Phe values of the DBS samples stored at 4℃ compared with the original measured values between 2007-2011,he results shows no significant difference,P〉0.05. The DBS stored in 4℃ within 1-2 years,the TSH values detected by conventional method compared with the original values,the results shows no significant difference,P〉0.05,and that stored for 3-5 years,the results shows significantly different,P〈0.01,and we use the time-delay method detected the DBS stored for 3-5 years,the results also shows significantly different. P〈0.01.The TSH values detected by the time-delay method also have significantly different with the original values,P〈0.01. Conclution:Dried blood filter cards stored at 4℃ within 1 to 5 years,the Phe detected values have no obvious relationship with the save time and the elution time,and also no significant differences with the original values.The DBS stored within 1-2 years,the TSH values were stable,and that stored of 3-5 years,the TSH values have significant differences with the original values,and the time-delay method can not make the TSH values restored to its original levels.
