异牡荆苷-6″-油酸酯和异荭草苷-6″-油酸酯合成及抗氧化性研究

摘  要:研究了固定化脂肪酶B(Novozmy 435)催化竹叶黄酮粗品与不饱和脂肪酸油酸的酰化反应,经液液萃取,薄层层析,硅胶柱层析分离纯化,得到两种酰化物单体;采用ESI-MS,~1H NMR及^(13)C NMR对目标产物的结构进行表征,确定产物分别为异牡荆苷-6″-油酸酯和异荭草苷-6″-油酸酯。讨论了酰化产物的抗氧化活性,包括葵花籽油中的氧化诱导时间、还原力、清除DPPH自由基的能力和清除羟基自由基的能力。结果表明:酰化产物在葵花籽油中的氧化诱导时间延长;还原能力、清除DPPH自由基和羟基自由基的能力在试验测试范围内均有下降。Immobilised Candida antarctica lipase B(Novozmy 435) was used to acylate flavonoid glycosides from bamboo leaves with oleic acid as acyl donors. After Liquid-liquid extraction, the products of esterification reactions were subjected to thin-layer chromatography(TLC), a silica gel column(700 mm × 35 mm); Then two types of acylated flavonoids were obtained. The purified products were analyzed by ESI-MS, ~1H-NMR and^(13)C-NMR and the two types of acylated flavonoids were isoorientin-6″-oleate and isovitexin-6″-oleate. The antioxidant activity of acylated products were evaluated through different antioxidant tests, including induction time values in sunflower oil, reducing capacity, DPPH radical scavenging activity and hydroxyl radical scavenging activity. The results showed that the induction times of easters increased; reducing power, radical scavenging activity against DPPH and hydroxyl radical decreased in the experimental test range.
