摘  要:为了制定蛋白酶K酶活力测定方法标准提供理论依据,研究了不同底物对蛋白酶K酶活力测定的影响。分别以6种常见蛋白质为底物,5种有代表性蛋白酶为对照,结果表明:所有受试蛋白酶均在以酪蛋白为底物时,酶活力最高。以角蛋白为底物时,蛋白酶K的相对酶活力(33.97%)低于碱性蛋白酶(58.34%)和胰蛋白酶(48.46%),远远高于菠萝蛋白酶(-2.92%)和木瓜蛋白酶(2.84%)。以血红蛋白为底物时,蛋白酶K的相对酶活力(25.70%)仅次于碱性蛋白酶(50.95%),远远高于胰凝乳蛋白酶(5.38%)和菠萝蛋白酶(7.61%)。以牛血清白蛋白、组蛋白和明胶为底物时,蛋白酶K酶活力很低甚至检测不到。因此,建议选择血红蛋白作为测定蛋白酶K酶活力的底物。In this paper,the effect of different kinds of substrates on determination of proteinase K activity was studied to provide a theoretical basis for the development of a method standard for the determination of proteinase K activity.Six kinds of common proteins were used as substrates,and five kinds of representative proteases were used as the controls,respectively.The results showed that the activity of all kinds of proteinase tested was the highest with casein as substrate.Using keratin as substrate,the relative activity of proteinase K(33.97%)was lower than that of alkaline proteinase(58.34%)and trypsin(48.46%),and far higher than that of bromelain(-2.92%)and papain(2.84%).Using hemoglobin as substrate,the relative activity of proteinase K(25.70%)was lower than that of alkaline proteinase(50.95%),and much higher than that of chymotrypsin(5.38%)and bromelain(7.61%).The activity of proteinase K was extremely low or could not be detected with BSA,histone and gelatin as the substrates.Therefore,hemoglobin was suggested to be as the substrate for determining proteinase K activity.
