摘 要:利用超声波辅助提取草鱼皮胶原蛋白,并通过SDS-PAGE、圆二色谱法对超声作用得到的草鱼皮胶原蛋白结构和变性温度等进行了研究。通过响应面试验得到最优工艺条件:超声功率310 W,超声时间10 min,酶解时间29 h,提取率达到68.67%。SDS-PAGE和CD显示:胶原蛋白为典型Ⅰ型,且纯度较高,变性温度为36℃,胶原蛋白的三螺旋结构完整,超声波对其结构无明显影响,且具有生物活性。The ultrasonic-assisted solvent extraction of grass carp skin' s collagen was carried out, and the gained collagen' s structure and denaturing temperature were studied by SDS-PAGE and circular dichroism spectrocopy. The experiments by response surface methodology showed that the optimal processing conditions were 310-W ultrasonic power,10-min ultrasonication and 29-h enzymolysis ,in which the extraction rate reached 68.67%. The research by SDS-PAGE and circular dichroism speetroeopy indicated that the collagen was of Type I and had a comparatively high purity, its denaturing temperature was 36℃, and its triple-helix structure was intact and not influenced by ultrasonic wave, and also had a biological activity.
