摘  要:为优化双孢蘑菇酶解的最佳工艺条件,以水解度为指标,研究了酶的种类、酶解时间、料液比、pH、酶添加量和酶解温度对双孢蘑菇的水解度的影响。在单因素试验的基础上,通过响应面法对双孢蘑菇酶解工艺进行优化。结果表明:采用复合蛋白酶,最佳酶解条件为酶添加量1%(w:w)、料液比1:20(w:v)、温度50℃、时间8 h、pH8.0,在此条件下,双孢蘑菇酶解液的水解度达到(31.90±0.13)%,游离氨基酸总量约是酶解前的1.3倍,呈味氨基酸含量是酶解前的1.5倍。研究结果为双孢蘑菇的高值化利用及附加产品的开发提供了新的思路。To optimize technological conditions for enzymatic hydrolysis of Agaricus bisporus, the effect of enzymic species, hydrolysis time, material/liquid ratio, pH, enzyme dosage and temperature on the degree of hydrolysis (DH) of Agaricus bisporus were studied. Based on the single experiment, response surface method was employed to optimize conditions for the enzymatic hydrolysis of Agaricus bisporus. The optimal hydrolysis conditions were found as follows: enzyme dosage 1% (w:w), material/liquid ratio 1:20 (w:w), temperature 50 ℃, time 8 h, pH 8.0. Under this conditions, the DH of enzymatic hydrolysate of Agaricus bisporus was (31.90±0.13)%, and the total content of free amino acids of enzymatic hydrolyzate was increased to 1.3 times after hydrolysis. The results provided a reference for deep-processing and utilization of Agaricus bisporus.
