摘 要:目的:研究高生物产量与糖含量的葡萄藻培养技术,探讨其多糖的体内抗菌活性。方法:采用多次划线分离获得葡萄藻单一藻种,选用BG11培养基研究其在不同光照强度和培养温度条件下的生长及糖含量的变化。以秀丽隐杆线虫为模式动物,研究多糖对金黄色葡萄球菌和沙门氏菌的抗菌效果。结果:分离获得的葡萄藻在光照强度65μmol·m^(-2)·s^(-1),培养温度28℃的条件下,培养14 d时其生物量能够达到0.92 g/L(DW);在光照强度65μmol·m^(-2)·s^(-1),培养温度33℃的条件下,其稳定期糖含量最高,达到干重的35.99%。葡萄藻多糖对秀丽隐杆线虫染毒的Gram+与Gram-均具有显著的抑菌效果(P<0.05),在其使用浓度等于或低于抗生素浓度时,其抑菌效果优于抗生素。结论:通过培养条件优化获得的葡萄藻多糖具有较好的抗菌能力。Objective: To study the cultivation technology of high biological yield and sugar content of Botryococcus sp., and the antibacterial activity of the polysaccharides from Botryococcus sp. Methods: The single Botryococcus sp. was obtained by repeatedly crossed isolation, and the trends of biomass and total sugar contain of this Chlorella were studied under different light intensity and incubation temperature with BGll culture medium. The antibacterial activity of the Botryococcus polysaccharide was investigated further by the Staphylococcus and Salmonella experiment of Caenorhabditis elegans in vivo. Results: The biomass of Botryococcus could reach 0.92g/L(DW) under the cultural condition of the light intensity of 65 μmol.m^-2.s^-1, incubation temperature of 28℃ at the 14 th day, and the sugar content of Botryococcus could reach 35.99%(DW) under the same light intensity and the incubation temperature of 33℃. The polysaccharide from Botryococcus has significant inhibitory effect on Caenorhabditis elegans exposure Gram+ and Gram (P〈0.05). In its use of concentration is equal to or lower than the concentrations of antibiotics, the effect the polysaccharide is better than the antibiotics. Conclusion: The polysaccharide from Botryococcus obtained through the optimization of culture conditions had an excellent antibacterial ability.
