摘 要:通过Plackett-Burman试验设计和响应面法优化粗壮脉纹孢菌固态发酵农副产品产类胡萝卜素的工艺条件,并对类胡萝卜素抗氧化能力进行研究。结果表明,固态发酵农副产品产类胡萝卜素的最佳工艺条件为:米糠、甘薯渣和啤酒糟的添加比例分别以34.25%,45.75%和20%,硫酸铵0.51%,初始含水量72.15%,接种量1%,发酵时间120 h,p H 6.5。在此条件下类胡萝卜素的产量达966.21μg/g培养物。粗壮脉纹孢菌所产类胡萝卜素对DPPH自由基、超氧自由基、羟基自由基均有较强的清除能力,比同浓度的番茄红素、β-胡萝卜素清除自由基能力强。此外,还具有较高的总抗氧化能力和较强的金属螯合能力。本研究为类胡萝卜素的开发利用提供了理论基础。In this paper, the conditions of solid-state fermentation for producing carotenoids were optimized with agricultural by-products by Neurospora crassa, through Plackett-Burman experimental design and response surface method, and the antioxidant capacity of carotenoids were studied. The carotenoids production was 966.21μg/g when the fermenta- tion were carried out on the optimized medium, which composed of 0.51% ammonium sulphate, 20% brewer's grain, 34.25% potato residues, 45.75% rice bran (calculated in dry weight) and the initial moisture content is 72.15%, pH 6.5 for 120 h. The results of antioxidant experiments indicated that: compared with common antioxidant, lycopene and β-carotene, the scavenging capability of carotenoids produced by Neurospora crassa towards DPPH, superoxide and hydroxyl radical were stronger in the experimental concentration. In addition, carotenoids produced by Neurospora crassa exhibited high total antioxidant capability and good chelating ability of metals.
