摘 要:目的建立固相萃取-高效液相色谱方法测定食品中米酵菌酸含量。方法样品经氨水-甲醇超声提取,上清液经固相萃取柱净化后,进高效液相色谱仪采用C18色谱柱分离,检测波长267 nm,外标法定量。结果米酵菌酸在0.3~40.0μg/ml质量浓度范围内线性关系良好,为0.999 4,加标回收率在94.6%~104.4%之间,相对标准偏差均<3.0%,检出限为0.005 mg/kg。结论该方法具有灵敏度高、准确度高、样品前处理简单等优点,可满足食品中米酵菌酸含量的检测要求。Objective To establish a solid extraction-high performance liquid chromatography method for the determination of bongkrekic acid in food. Methods Bongkrekic acid in samples were extracted by ammonia-methanol,and the supernatant was cleaned up by solid phase extraction column. Target compound was separated on a C18 column and quantified by external standard method with the wavelength of 267 nm. Results The calibration curve for bongkrekic acid was linear in the range of 0. 3-40 μg/ml with the correlation coefficient of 0. 999 4,and the spike recoveries were in the range of 94. 6%-104. 4% with relative standard deviation lower than 3. 0 %. The detection limit was 0. 005 mg/kg. Conclusion The method has the advantages of high sensitivity,good accuracy,rapid determination,and can be suitable for detection of bongkrekic acid in food.
