摘 要:以香菇柄为原料,采用超声-闪式联合方法提取麦角甾醇,探讨提取时间、闪提转速、液料比、超声功率对麦角甾醇提取的影响。在单因素试验的基础上,通过响应面法优化超声-闪式联合提取香菇柄中麦角甾醇的工艺参数,利用S180荷瘤小鼠模型和酶联免疫吸附试验(ELISA)研究麦角甾醇的抗肿瘤活性及其机制。结果表明,利用超声-闪式联合法提取香菇柄中麦角甾醇的优化工艺参数为:室温条件提取14 min,闪提转速10 000 r/min,液料比36 m L/g,超声功率450 W,此条件下香菇柄中麦角甾醇的提取率为2.818 mg/g。可见超声-闪式联合法提取时间短,提取率较高;麦角甾醇高【400 mg/(kg·d)】、中【300 mg/(kg·d)】、低【(200 mg/(kg·d)】剂量组的瘤重抑制率分别为41.64%,29.90%,19.52%,与模型对照组相比均有显著性差异(P<0.05),小鼠血管内皮生长因子(VEGF)A的表达水平也较模型组显著降低(P<0.05),说明香菇柄麦角甾醇具有抗肿瘤活性,其抑瘤机制可能与抑制VEGF-A生成有关。Ultrasonic combined with flash extraction was used to extract the ergosterol from Lentinula edodes stipe. The extraction time, the flash extraction speed, the liquid-material ratio and ultrasonic power are taken as the affect factors. On the basis of single-factor test, the response surface methodology is used to optimize extraction condition. In vivo antitumor activity of ergosterol from Lentinula edodes stipe was tested by mouse Slso transplant tumor model and ELISA. The results showed that optimization of process parameters with ultrasound-flash extraction ergosterol from Lentin- ula edodes stipe were as follow: room extraction 14 min, flash extraction speed 10 000 r/rain, liquid-material ratio 36 mL/g, ultrasonic power 450 W. Under the optimal process conditions, the extraction yield of ergosterol from Lentinula e- dodes stipe is 2.818 mg/g. So ultrasound-flash extraction method is good to shorten extraction time and improve extraction yield. The tumor weight inhibition rate of ergosterol at doses of 400, 300 and 200 mg/(kg.d) were respectively 41.64%, 29.90%, 19.52%. Compared with the model control group, there were significant differences (P〈0.05). The vascular en- dothelial growth factor (VEGF) A of mouse was also significantly lower than the model control group (P〈0.05). There- fore, ergosterol from Lentinula edodes stipe have anti-tumor activity, and the inhibitory mechanism may be related to in- hibition of VEGF-A generated.
