摘 要:目的:探究鲫鱼卵唾液酸糖蛋白的食用安全性及其在体内的蓄积靶器官。方法:采用急性毒性试验,Ames试验、骨髓微核试验、小鼠精子畸形试验等3项遗传毒性试验和大鼠90 d喂养试验对鲫鱼卵唾液酸糖蛋白进行毒理学评价;以体内唾液酸含量变化为追踪目标,ICR小鼠被连续灌胃鲫鱼卵唾液酸糖蛋白10 d后,按试剂盒法检测其脾、心、肾、肝、肌肉、脑、睾丸、卵巢及骨中唾液酸含量。结果:小鼠经口最大耐受剂量大于45 g/(kg bw),鲫鱼卵唾液酸糖蛋白属无毒级别,3项遗传毒性试验结果均为阴性,90 d喂养试验未见大鼠脏体比、血常规、血清及尿液生化指标出现明显异常;经鲫鱼卵唾液酸糖蛋白干预后,小鼠股骨、胫骨及腰椎中唾液酸含量显著上升(P<0.01),其它器官唾液酸含量无显著变化。结论:鲫鱼卵唾液酸糖蛋白无急性毒性、遗传毒性及亚慢性毒性,骨组织为其蓄积靶器官。Objective: To investigate the food safety and accumulative target organs in mice gavaged with sialyglyco- proteins from crucian carp carassius auratus eggs (Ca-SGP). Method: The toxicological evaluation of Ca-SGP were con- ducted by using methods of acute toxicity test, three terms of genetic toxicity tests including Ames test, bone marrow cell micronucleus test and mice sperm abnormality test, and ninety feeding study. With the changes of sialic acid content in mice as the tracking target, after ICR mice were gavaged for 10 days continuously, the sialic acid contents in various organs were determined. Results: The maximum tolerated dose of mice were greater than 45 000 mg/kg.bw, suggesting that Ca-SGP was non-toxic. The genetic toxicity tests were all negative. The targets of organ/body weight ratio, peripheral hemogram, serum and urine biochemistry indices between 90-day fed rat in experimental group and control groupwere no significant difference The sialic acid contents were markedly increased in femur, tibia, and lumbar in mice after treated by Ca-SGP (P〈O.01), while there were no remarkable changes in other orgens. Conclusion: Ca-SGP had no acute toxi- city, genetic toxicity and subchronic toxicity under our evaluation system, and its accumulative target organ was bone tissue.
