摘 要:目的:从土壤中筛选能够产生中性耐热β-葡萄糖苷酶的芽孢杆菌菌株,并对该菌株进行鉴定。方法:将土壤样品处理后涂布于七叶苷平板上,37℃培养2 d,挑选能够产生黑褐色水解圈的菌株,划线分离,得到单菌落。用p NPG作为底物进行酶活复筛,选取酶活性最高的菌株,并通过菌落形态、生理生化特征和分子生物学方法对其进行鉴定。结果:从土壤样品中筛选得到20株能够产β-葡萄糖苷酶的菌株,经酶活复筛得到1株酶活性最高的菌株,命名为ZJ1308,其酶活为0.46 U/m L。通过鉴定该菌株为蜡状芽孢杆菌。结论:该菌株的获得为β-葡萄糖苷酶的产业化应用提供了良好基础。Objective: To screen and indentify a neutral and heat-tolerant /^-glucosidase-producing Bacillus strain from the soil. Methods: The soil samples were treated and streaked onto the agar-plate containing esculin. After the samples were cultured at 37℃ for 48 h, the colonies that can produce larger dark brown hydrolysis cycle were picked and isolated. The strain with the highest β--glucosidase activity was re-screened with pNPG as the substrate, and was i- dentified by the colony morphology in combination with the physiological and biochemical characteristics and the molecu- lar biology technology. Results: 20 strains that could produce/3-glucosidase were obtained from soil samples. The strain with the highest β-glucosidase activity was obtained by re-screening, and was named ZJ1308 and identified as Bacillus cereus. Its β-glucosidase activity was up to 0.46 U/mL. Conclusion: This study lays a good foundation for the industrial application of β-glucosidase.
