摘  要:β-葡萄糖苷酶是纤维素分解酶系中的重要组成部分,具有广泛的应用前景。浓香型大曲因其独特的制作工艺,其微生物含量非常丰富。以浓香型大曲为菌源,分离纯化出一株高产β-葡萄糖苷酶的菌株,经鉴定该菌株属于枯草芽孢杆菌(Bacillus subtilis)。单因素实验结果表明在含1%水杨苷发酵液体培养基中,最佳产酶培养时间72 h、生长温度40℃、生长初始p H为5.5。为了提高产酶能力,采用正交实验对产酶条件进行了优化,获得最佳产酶条件。在此条件下,该菌株发酵液β-葡萄糖苷酶活力可达到55.7 U/m L。研究通过对大曲中产β-葡萄糖苷酶微生物的筛选,获得一株高产菌株,为从大曲中分离产β-葡萄糖苷酶微生物提供了方法,拓展了产纤维素酶微生物的菌种资源;通过对高产菌株最适产酶条件和产酶能力的研究,为进一步开发该菌株奠定了一定的基础。The β-glucosidase is an important component of cellulose, with broad application prospect. Luzhou-flavor Daqu(yeast for making hard liquor) is rich in microbial species with special process. In this study, we isolated a strain that could high secretion β-glucosidase from Luzhou-flavor Daqu, and the strain was identified as Bacillus subtilis. The results of single factor experiments showed that the optimal culture time was 72 h, the optimum growth temperature was 40 ℃, and the optimum initial p H was 5.5 in the medium containing 1% salicin. In order to improve the ability of producing enzyme, orthogonal experiment was used to optimize the conditions of enzyme production, and the optimum conditions were obtained. In this condition, the activity of β-glucosidase in the fermentation broth could reach 55.73 U/m L. In this study, a β-glucosidase high secretion strain was isolated from Daqu, it could provide a method for isolating strain that could secretion β-glucosidase and expanded the micro bial resources that could produce cellulose, and laid a foundation for further developed the strain through optimum the conditions of enzyme production and ability of produce enzymes.
