摘 要:以重庆市特色蔬菜胭脂萝卜为原材料提取花青素,以吸附率和解析率为指标考察6种大孔树脂对胭脂萝卜花青素的纯化效果,从中筛选出最佳大孔树脂并探讨其吸附性能和纯化工艺,分析最佳树脂纯化前后胭脂萝卜花青素的色价及抗氧化活性。结果表明,AB-8树脂在吸附和解析3h后其吸附率和解析率达到91.2%和96.6%。AB-8树脂的纯化工艺为:上样浓度为0.80 mg/mL,上样液pH 4,上样流速8 mL/min,洗脱流速6 mL/min。经AB-8树脂纯化后的胭脂萝卜花青素色价是粗提液的9倍。0.75mg/mL的胭脂萝卜花青素纯化液抗氧化活性显著高于同浓度的花青素粗提液及维生素C,经AB-8树脂纯化的胭脂萝卜花青素纯化液对羟自由基、DPPH自由基及油脂的清除能力显著提高。以上结果表明,AB-8树脂是胭脂萝卜花青素的最佳纯化树脂。Anthocyaninsextracted from carmine radish, a regional vegetable in Chongqing, was used as the raw material.Purificationeffects of six resins was studied through adsorption and desorption rate, and adsorption effect and purification technologyof optimum resin were discussed.Meanwhile, color value and antioxidant activity of carmine radish anthocyanins before and after purificationwere analyzed.The results showedthe adsorption rate and desorption rate of AB-8 resinwere 91.2% and 96.6% after 3h. The optimized conditions of AB-8 were sample concentration 0.80 mg/mL, pH value of 5, thesample flow rate of 8 mL/min, and the elution flow rate of 6 mL/min. The color value was increased about 9 times afterAB-8 resin purification.The antioxidant activity of 0.75 mg/mLpurified anthocyanins was significantly higher than the same concentration of extrude anthocyanins and vitamin C.The scavenging activity ofhydroxyl radical, DPPH free radical and grease were enhanced after AB-8 resin purification. AB-8 resin was the best separating and purifying resin of carmine radish anthocyanins.
