摘 要:鼠李糖脂是一种生物表面活性剂,能被生物完全降解,广泛用于石油工业、农业、生态环境。针对一般菌种产量低、生产成本高的问题,本实验对产鼠李糖脂铜绿假单胞菌株进行诱变育种和发酵条件研究,获得一株鼠李糖脂产量达8.14g/L高产突变菌株S19-4,通过单因素和正交试验得到菌株产鼠李糖脂合适培养基组成分为(g/L):NaNO_3 4.5,KH_2PO_4 0.90,K_2HPO_4 0.90,MgSO_4 2.0,MnSO_40.05,FeSO_4 0.04,酵母膏1.5,玉米油65.0;最佳发酵条件:接种量3%、初始p H值7.1-7.2、装液量80m L/500m L瓶、转数210r/min、温度35℃、发酵时间72h。最终鼠李糖脂平均产量为38.9g/L。Rhamnolipidis a kind ofbiosurfactants that can be completely degraded by the organism. It iswidely used in petroleum industry, agriculture and ecological environment. To solve the problems of low yield and high pro- duction cost,the screening of the rhamnolipid-producing mutant strains and optimization of fermentation conditions were studied. After the mutagenesis, the rhamnolipid yield of mutant strain S19-4 could reach 8.14g/L. Using sin- gle factor experiment and orthogonal experiment got the optimum medium formulas(g/L) that was as follows: NANO3 4.5, KH2PO4 0.90, K2HPO4 0.90, MgSO4 2.0, MnSO4 0.05, FeSO4 0.04, yeast extract 1.5, corn oil 65.0. The optimum inoculation amount was 3%, the optimum initial pH value was between 7.1-7.2, the optimum media volume was 80mL, the optimum revolutions was 210r/rain, the optimum temperature was 35℃, the optimum fer-mentation time was 72h. Finally confirmed, the highest average rhamnolipid production of Pseudomonas aeruginosa S19-4 was 38.9g/L.
