摘  要:目的:用4种不同的干燥提取方法从香菇柄中提取活性多糖,对比4种方法得到的多糖的体外抗氧化的差异及贮存时间对于抗氧化的影响。方法:根据提取方法的不同将所得香菇多糖命名为:冷冻干燥多糖(FDP)、醇沉-冷冻干燥多糖(AFDP)、喷雾干燥多糖(SDP)和醇沉-喷雾干燥多糖(ASDP)。评价了香菇多糖的体外抗氧化活性:对ABTS自由基、DPPH自由基的清除能力以及对Fe3+的还原力。进一步探究了不同保存时间对香菇多糖上述活性的影响。最后以保存时间(提取后于-20℃保存10,20,30,40天)为单一变量,研究抗氧化活性的变化。结果:相同多糖浓度下,冷冻干燥得到的多糖的体外抗氧化活性高于喷雾得到的多糖,醇沉得到的多糖的体外抗氧化活性要低于没有醇沉得到的多糖。所以选择了无醇沉冷冻干燥的多糖来研究时间对于体外抗氧化活力的影响,并且香菇柄多糖的抗氧化活性呈现剂量和时间依赖性。结论:香菇柄多糖的最优提取方法为冷冻干燥法,在考虑药效和经济成本平衡的前提下,醇沉冷冻干燥得到的多糖为好。Objective:Four kinds of drying extraction methods are used to extract the active polysaccharides from the stems of mushrooms.The antioxidant differences of the polysaccharides obtained from the four methods and the storage time are compared.Methods:Named the lentinan by extraction methods as follows:freeze drying polysaccharide(FDP),alcohol precipitation-freeze drying polysaccharide(AFDP),spray-drying polysaccharide(SDP)and alcohol precipitation-spray dring polysaccharide(ASDP).The antioxidant activity of lentinan is evaluated in vitro:the scavenging ability on ABTS free radical,DPPH free radical and reducing ability of Fe3+.To further explore the effect of different preservation time on the activity of lentinan above,the experiment is repeated at four different storage time on day 10,day 20,day 30 and day 40 under -20 ℃.Results:The antioxidant activity of the polysaccharides obtained by freeze drying is higher than that of the polysaccharides obtained by spraying,and the antioxidant activity of non-alcohol precipitated polysaccharides is higher than that of alcohol-precipitated polysaccharides.The antioxidant activities of the polysaccharides from Lentinus edodes are time-dependent.The inhibitory rate increases in line with the concentration.Conclusion:The polysaccharides obtained by freeze-drying without alcohol precipitation have the highest and longest antioxidant activity in vitro.Under the premise of considering the balance of efficacy and economic cost,the polysaccharides obtained by freeze-drying alcohol precipitation are the best choice.
