摘 要:以河蚬为原料,采用Alcalase、Neutrase、Flavourzyme、Papain四种蛋白酶水解河蚬,考察不同酶酶解产物的抗氧化活性包括DPPH自由基清除能力、超氧阴离子自由基(O-2·)清除能力、羟自由基(·OH)清除能力及还原力。研究发现,Alcalase2 h酶解产物DPPH自由基清除能力最佳,清除率为83.17%,Alcalase8 h酶解产物O-2·的清除率最高达到72.52%。Papain6 h酶解产物·OH清除效果最佳,清除率达83.86%,Papain 6 h酶解产物还原力最高,酶解产物OD700nm值为0.74。可见蛋白酶种类和酶解时间对河蚬酶解产物的抗氧化活性具有较大影响,且抗氧化活性指标不一样,不同的蛋白酶酶解河蚬产物的抗氧化活性也有较大差异。Four kinds of protease,Alcalase,Neutrase,Flavourzyme and Papain,were used for the hydrolysis of freshwater clam,and antioxidant activities of hydrolysates in terms of 1,1-diphenyl-2-picrylhydrazyl( DP PH) radical scavenging activity,superoxide radical anion( O2^-·) scavenging activity,hydroxyl radical( ·OH) scavenging activity and reducing power were evaluated. The results showed that hydrolysate produced by Alcalase at 2 h exhibited the best DPPH radical scavenging ability and its scavenging activity reached 83. 17%.Hydrolysate produced by Alcalase at 8 h showed the highest O2^-· scavenging activity and it was 72. 52%. Hydrolysate produced by Papain at 6 h exhibited the best · OH scavenging ability and its scavenging activity reached 83. 86%. Hydrolysate produced by Papain at 6 h displayed highest reducing power and its OD700 nm value was 0. 74. The results showed that the protease type and hydrolysis time had significant effects on the antioxidant activity of the hydrolysate. The antioxidant activity of hydrolysates produced by different protease were different from each other using the different indicators of antioxidant activity. Overall,these results could provide theoretical basis for preparation and use of freshwater clam hydrolysate with antioxidant activity.
