摘 要:本实验以多形汉逊酵母(Hansenula polymorpha,H.polymorpha)无机硒转化率及其胞内谷胱甘肽(Glutathione,GSH)产量为评价指标,通过单因素实验、正交试验研究亚硒酸钠(Na_2SeO_3)添加量、添加时间、接种量等参数,确定制备富硒-GSH多形汉逊酵母的最佳发酵工艺,利用MTT比色法研究制备的富硒-GSH多形汉逊酵母对大鼠的抗氧化活性。结果表明,在4%接种量、37℃培养温度、120 r/min转速和发酵培养12 h后(48 h发酵周期)添加终浓度为30μg/m L Na_2SeO_3的条件下,多形汉逊酵母无机硒转化率及其胞内GSH产量为97.28%±1.89%和(215.21±3.01)mg/L。同时,与出发多形汉逊酵母组相比,饲喂富硒-GSH多形汉逊酵母组的SD大鼠的血清和肝脏中,GSH水平分别提高39.5%(p<0.01)、45.2%(p<0.001),T-AOC提高46.3%(p<0.01)、45.2%(p<0.05),GSH-Px提高48.3%(p<0.001)、52.3%(p<0.01),SOD提高68.4%(p<0.01)、45.1%(p<0.05),CAT提高39.5%(p<0.01)、49.6%(p<0.001),MDA降低21.3%(p<0.05)、38.5%(p<0.01)。表明富硒-GSH多形汉逊酵母可显著提高大鼠的抗氧化活性。研究结果为利用多形汉逊酵母高效、低成本协同制备有机硒和GSH提供新思路、新途径,并为其在保健食品开发领域的应用提供技术支撑。In this experiment,the inorganic selenium conversion rate of Hansenula polymorpha(H.polymorpha)and the yield of intracellular glutathione(GSH)were used as evaluation indexes.The effects of parameters such as sodium selenite(Na2SeO3)addition,addition time,inoculum size were studied by single factor experiment and orthogonal experiment to determine the optimal fermentation process for preparing selenium-glutathione-enriched H.polymorpha.The physiological function of selenium-glutathione-enriched H.polymorpha in rats was analyzed by MTT colorimetric assay.The optimal condition of inorganic selenium produced by H.polymorpha was following:inoculation amount of 4%,Na2SeO3with a final concentration of 30μg/mL addition to the fermentation medium after 12 h cultivation,the fermentation period of 48 h,the culture temperature of 37℃and the shaking speed of 120 r/min.Under this condition Na2SeO3conversion rate and intracellular GSH yield of H.polymorpha reached 97.28%±1.89%and(215.21±3.01)mg/L,respectively.Meanwhile,the serum and liver GSH levels of SD rats in the selenium-glutathione-enriched H.polymorpha group were increased by 39.5%(p〈0.01)and 45.2%(p〈0.001),T-AOC with the increase of 46.3%(p〈0.01),45.2%(p〈0.05),GSH-Px with the increase of 48.3%(p〈0.001),52.3%(p〈0.01),SOD with the increase of 68.4%(p〈0.01),45.1%(p〈0.05),CAT with the increase of 39.5%(p〈0.01),49.6%(p〈0.001),and MDA with the decrease of 21.3%(p〈0.05),38.5%(p〈0.01),respectively,which indicated that selenium-glutathioneenriched H.polymorpha could significantly increase the antioxidant activity of rats.The results would provide novelty pathways for preparing inorganic selenium and glutathione by H.polymorpha and offered a technical support for its application in food development.
