摘  要:观察S-层蛋白介导的嗜酸乳杆菌对溃疡性结肠炎的治疗效果,探讨嗜酸乳杆菌对肠黏膜细胞凋亡进程及大鼠血清中细胞因子表达水平的影响。随机将大鼠(n=75)均分为5组,分别为正常组、模型组、美沙拉嗪对照组、嗜酸乳杆菌干预组Ⅰ(有S-层蛋白菌)和嗜酸乳杆菌干预组Ⅱ(去S-层蛋白菌)。采用5%2,4,6-三硝基苯磺酸(TNBS)与50%乙醇混合液(1∶1)诱导,建立大鼠溃疡性结肠炎模型。造模2 d后,按照不同分组喂食相应药物,于造模后第8天处死所有大鼠,观察并测定下列指标:组织病理学特征、细胞凋亡率、Caspase-3与Caspase-8表达量、细胞因子(TNF-α、IL-6、IL-8)含量。相对于模型组的各指标,美沙拉嗪对照组、去除与未去除S-层蛋白的嗜酸乳杆菌干预组的各指标均有不同程度的下降(P<0.01,P<0.05),且未去除S-层蛋白的嗜酸乳杆菌比去除S-层蛋白的嗜酸乳杆菌具有更显著的疗效(P<0.01),说明嗜酸乳杆菌可以通过调节肠道免疫来缓解炎症,且S-层蛋白在此过程中发挥了重要作用。This study was to observe the intervention effects of Lactobacillus acidophilus on ulcerative colitis(UC)in mice mediated by Surface layer proteins through the influence of L. acidophilus on apoptosis of intestinal epithelium cells(IEC) and cytokines level in rat serum. Rats were divided into 5 groups on average, including normal group, model group, mesalazine group, L. acidophilus intervention group Ⅰ, L. acidophilus intervention group Ⅱ. Use 5% 2,4,6-TNBS/alcohol(1∶1) to establish UC model. Feed corresponding drugs to each group from the third day and kill all rats at the eighth day. Observe and detect indexes as follows: histopathologic characteristics, IEC apoptosis rate, expression quantity of cell apoptosis key enzymes Caspase-3 and Caspase-8, cytokines(TNF-α, IL-6, IL-8) in rat serum. Compared with the model group rats, the indexes of mesalazine group, L. acidophilus intervention group Ⅰand L. acidophilus intervention group Ⅱ had different degrees of decrease(P〈0.01,P〈0.05). Furthermore, efficacy of L. acidophilus with Slayer protein was more obvious than that without S-layer protein in curing UC(P 〈0.01). Results proved that L. acidophilus could relieve UC by regulating the intestinal immune, in which S-layer protein played an important role.
