摘 要:目的:建立定量检测雨生红球藻及其萃取物中虾青素含量的方法。方法:使用新型脂肪酶AstaE-H催化水解虾青素酯,再采用外标法在C30-HPLC上对游离态虾青素进行定量。结果:测定结果的RSD值达到2.63%,加样回收率为100.08%,检测下限为40ng/mL,线性范围为0.04~25μg/mL。结论:应用AstaE-H脂肪酶的方法具有重复性好、灵敏度和回收率高、反应副产物(如虾红素和顺式异构体)少等优点,优于国标GB/T31520-2015方法,且成本远低于美国药典方法,可在相关产业中推广应用。Abstract : Objective : This study aims to develop a method for the quantification of astaxanthin content from Haematococcus pluvialis extracts. Method .. Astaxanthin esters from alga were hydrolyzed by a novel lipase catalysis of Asta-E-H. The astaxanthin was then quantified on C30-HPLC by external standard. Results ~ RSD of the method was 2.63% and the recovery rate was 100.08%. The detection limit was 40ng/mL, linear range was 0.04 - 25mg/ mL. Conclusion ~ The method with Asta-E-H has the advantages on satisfied reproducibility, high sensitivity and good recovery rate, and less by-products (eg. astacene and Z-isomer) compared with those of national standard GB/T31520-2015 ; the method costs less than U.S pharmacopoeia method. The method has potential to apply in the related fields.
