摘 要:目的利用Soleris细菌快速检测仪器,以传统国标法菌落总数检测结果为校正数据集,建立快速检测冰淇淋菌落总数的Soleris曲线。方法样品溶于生理盐水,经均质混匀后,调节pH至6.7±0.2。用国标方法和Soleris仪器同时对样本进行检测,以国标法检测的菌落总数与Soleris检测响应时间建立和优化检测曲线和回归公式。结果得到的菌落总数检测回归校正曲线和回归校正公式为Y(lgCFU)=-0.9014DT+9.2182(r2=0.9575),用10个样品对回归公式进行的验证试验,准确性为90%;同时对Soleris检测结果的重复性进行验证,得到重现性为100%。结论改进后的检测方法具有时间短、效率高、结果准确的优势,可用于冰淇淋菌落总数的快速检测。Objective To establish a Soleris curve for rapid determination the total bacterial colony of ice cream by using Soleris rapid detection instrument, and the results of total bacterial colony detection by traditional national standard method were taken as the corrected data set. Methods The sample was dissolved in normal saline solution, and the pH was adjusted to 6.7±0.2 after homogenization. Samples were tested simultaneously with the national standard method and Soleris instrument, and the detection curve and regression formula were established and optimized with the total number of bacterial colonies detected by the national standard method and the response time of Soleris detection. Results The regression correction curve and regression correction formula obtained for total detection of total bacterial colony were Y(lgCFU)=-0.9014 DT+9.2182(r2=0.9575). Totally 10 samples were used to verify the regression formula, and the accuracy was 90%. At the same time, the repeatability of Soleris detection was verified, and the reproducibility was 100%. Conclusion This method has advantages of short time, high efficiency and accurate results, and can be used for rapid detection of total bacterial colony of ice cream.
