摘  要:研究乳源酪蛋白糖巨肽(casein glycomacropeptide,CGMP)对结肠癌细胞中的核转录因子(nucleartranscription factor,NF)-κB信号通路的作用,为其抗炎活性提供理论依据。本研究在前期探究"乳源酪蛋白糖巨肽对结肠癌HT-29细胞中COX-2、iNOS、GST-π表达的影响"的基础上,首先用脂多糖(lipopolysaccharide,LPS)刺激结肠癌HT-29细胞将NF-κB信号通路激活,采用免疫荧光法观测p65核移位来确定;随后用乳源CGMP干预激活NF-κB通路的HT-29细胞,用酶联免疫吸附剂测定细胞培养液中炎症因子和血管生成因子的表达水平,以探讨乳源CGMP消除结肠癌细胞活力的相关分子的变化。结果显示,1μg/mL的LPS能显著激活NF-κB信号通路,p65的核移位达到最佳水平;不同质量浓度的乳源CGMP均可不同程度地抑制HT-29细胞炎症因子和血管生成因子的表达水平。因此,乳源CGMP能够显著调节激活后的NF-κB信号通路,并有效抑制和改善结肠癌细胞的炎症状态,其机制为乳源CGMP通过有效抑制炎症因子和血管生成因子的表达,从而减缓结肠癌细胞的炎症程度。The effect of casein glycomacropeptide(CGMP)on the nuclear transcription factor(NF)-κB signaling pathway in colorectal carcinoma cells was assessed in order to provide a theoretical basis for its anti-inflammatory activity.As an extension of our previous work investigating the influence of CGMP on the expression of COX-2,iNOS and GST-πin colon cancer HT-29cells,this study determined whether the NF-κB signaling pathway in LPS challenged HT-29cells was activated through observing p65nuclear translocation by immunofluorescence microscopy.The challenged cells were treated with CGMP and the expression levels of inflammatory cytokines and angiogenesis factors in the cell cultures were detected by enzyme-linked immunosorbent assay(ELISA).The results showed that1μg/mL LPS significantly activated the NF-κB signaling pathway,and resulted in optimized nuclear translocation of p65.CGMP at all investigated concentrations could inhibit the expression levels of inflammatory factors and angiogenic factors in HT-29cells.Therefore,CGMP was capable of regulating the activated NF-κB signaling pathway and effectively restraining and improving the state of inflammation in colon cancer cells.This may be ascribed to inhibited expression of inflammatory cytokines and angiogenic factors,consequently reducing inflammation degree.
