摘 要:本文主要研究以芸豆蛋白为底物,分别用木瓜蛋白酶、中性蛋白酶、复合蛋白酶、胰蛋白酶和碱性蛋白酶在一定条件下进行酶解,以蛋白的水解度、抗氧化能力、对超氧阴离子的捕获能力为指标,选出3种最优酶。对3种酶进行复配,得到6种双酶组合,通过试验确定最佳的双酶水解组合。结果表明:碱性蛋白酶、胰蛋白酶和木瓜蛋白酶的酶解液具有较高的水解度、超氧阴离子清除率、抑制脂质过氧化的能力。双酶最佳组合为胰蛋白酶+碱性蛋白酶,水解度、超氧根清除率、抑制脂质过氧化的能力依次可达43.28%、37.27%、38.63%。可为后续的水解因素及水解工艺参数研究提供依据。This paper mainly studied the enzyme hydrolysis of kidney bean protein with papain,neutral protease, compound protease,trypsin and alkaline protease under certain conditions.Three optimal enzymes were selected based on the indexes of protein hydrolysis degree,antioxidant ability and superoxide anion capture ability.Six combinations of double enzymes were obtained by compounding three enzymes,and the best combination of double enzymes was determined through experiments.The results showed that the hydrolysates of alkaline protease,trypsin and papain had higher hydrolysis degree, superoxide anion scavenging rate and inhibiting lipid peroxidation.The best combination of double enzyme is trypsin + alkaline protease,and the degree of hydrolysis,superoxide root clearance rate and inhibition of lipid peroxidation can reach 43.28%,37.27% and 38.63% respectively.It can provide a basis for the subsequent study of hydrolysis factors and hydrolysis process parameters.
