摘  要:以金枪鱼红肉酶解液为研究对象,研究其在抗疲劳、抗氧化和肠道菌群调节方面的作用。本研究首先制备金枪鱼红肉酶解液,通过MALDI-TOF/TOF对多肽组成和丰度进行鉴定;随后对小鼠进行连续8周灌胃,评价其抗疲劳、抗氧化及对肠道菌群调节的能力。结果表明,金枪鱼红肉酶解液中富含与抗疲劳作用密切相关的精氨酸和亮氨酸、异亮氨酸、缬氨酸三种支链氨基酸。与对照组相比,酶解液分别延长了雌雄小鼠72.8%和70.8%的转棒时间。同时,雄性处理组血清中抗疲劳参数指标磷酸肌酸激(CK)酶活和乳酸(LD)含量分别降低12.1%和18.7%,乳酸脱氢酶(LDH)酶活增加29.8%;抗氧化参数指标GSH-Px和SOD分别增加38.4%和13.9%,MDA含量降低48.6%;而在雌性处理组中,CK酶活和LD含量分别降低3.7%和12.4%,LDH酶活增加13%,GSH-Px和SOD分别增加4.99%和1.74%,MDA含量降低39.6%,肝脏中各项指标的变化趋势与血清中一致。此外,酶解液降低了雌性小鼠肠道菌群物种丰富度和多样性,而只降低了雄性小鼠的菌群丰富度;拟杆菌门和厚壁菌门丰度在雌雄小鼠中呈现相反的变化趋势;乳酸菌、乳杆菌和短链脂肪酸产生菌等益生菌丰度上升;雌雄小鼠间仅存在3个共有关键OTUs。In this study,the anti-fatigue,anti-oxidation and modulating gut microbiota effects of enzymatic hydrolysate of tuna dark meat(EH-TDM)were investigated in mice.The EH-TDM was firstly prepared,the peptides components and abundance were measured via MALDI-TOF/TOF-MS,and the anti-fatigue,anti-oxidation and modulating gut microbiota effects were explored via EH-TDM treatment experiment for 8 weeks in mice.As shown in the results,firstly,most of the peptides contained arginine and branched-chain amino acids(leucine,isoleucine,valine),which showed anti-fatigue effects.Secondly,EH-TDM treatment increased the rolling stick time by 72.8%and 70.8%in male and female mice,respectively.At the same time,EHTDM treatment in male mice decreased the phosphocreatine(CK)activity and lactic acid(LD)contents in serum by 12.1%and 18.7%,and increased the activity of lactate dehydrogenase(LDH)by 29.8%,all these three indexes showed close relationships with anti-fatigue function.In addition,EH-TDM treatment also decreased the MDA contents in serum by 48.6%,and increased the activity of GSH-Px and SOD by 38.4%and 13.9%,which were involved in anti-oxidation function.On the other hand,EH-TDM treatment in female mice decreased the CK activity and LD content in serum by 3.7%and 12.4%,increased the activity of LDH by 13%,whereas increased the activity of GSH-Px and SOD by 4.99%and 1.74%,and decreased the MDA content by 39.6%.Otherwise,similar change trends of these indexes were observed in liver.Thirdly,EH-TDM treatment decreased the bacterial richness in both male and female mice,whereas only decreased the bacterial diversity in female mice.Moreover,different change profiles were observed in the phylum Bacteroidetes and Firmicutes in mice,and various benefit strains,such as Lactobacillus and short-chain fatty acid-producing bacteria,were increased after EH-TDM treatment,whereas only three key OTUs were detected in both male and female mice.
