摘  要:针对虾青素油生物利用度低的问题,本文比较研究了水分散型虾青素纳米粉剂(Ast-nano)、虾青素微囊剂(Ast-micro)和虾青素油(Ast-oil)对急性酒精氧化损伤小鼠的抗氧化作用。其中,Ast-nano以鲑鱼精DNA-壳聚糖为载体,利用大分子共组装法制得纳米粒,辅以冷冻干燥技术获得干燥粉体。利用高效液相色谱法、动态光散射法等检测了各虾青素制剂的含量、粒径和粒径分布等理化性质。通过建立小鼠急性酒精氧化损伤模型,对Ast-nano、Ast-micro和Ast-oil的体内抗氧化效果进行评价。研究表明,Ast-nano和Ast-micro中虾青素含量分别为18.34%和7.36%,与雨生红球藻中虾青素含量(5%)相比分别提高了266.8%和47.2%,且均可在30 s内实现快速溶解。50 nm的Ast-nano由于冻干保护剂的差异使其复溶后粒径控制在200~500 nm,但粒子的ζ电位均大于40 mV,有利于悬液的稳定。体内抗氧化研究发现,Ast-nano在提高小鼠体内GSH和SOD水平及降低体内MDA和PC含量能力方面显著优于Ast-micro和Ast-oil。研究结果表明,水分散型虾青素纳米粉剂具有比虾青素油更优的抗氧化活性。Micro-and nano-encapsulations technology as the core technologies of nanotechnologies have shown great potentials in the efficient transportation and absorption of bioactive compounds.Due to the low bioavailability of astaxanthin oil,a series of astaxanthin formulations including water-dispersible astaxanthin-encapsulated nanopowder(Ast-nano),astaxanthin microcapsules(Ast-micro)and astaxanthin oil(Ast-oil)were employed to study the effects of different formulations on the antioxidant activity(in vivo).As for Ast-nano,the formulation was prepared with macromolecular co-assembly method,and freeze-drying technology was finally used to obtain solid powders.The physical and chemical properties of each astaxanthin formulation were estimated with HPLC,dynamic light scattering(DLS)among others.The in vivo antioxidant activity of different astaxanthin formulations was evaluated with the acute alcohol oxidative damage mice model.The results showed that the astaxanthin content in Ast-nano and Ast-micro was 18.34%and 7.36%,respectively,which increased by 266.8%and 47.2%as compared with the astaxanthin content of H.pluvialis(5%).Both of the water-dispersible astaxanthin formulations could be rapidly dissolved within 30 s.Ast-nano with a particle size around 50 nm changed into a larger size(200~500 nm),which was caused by the cryoprotectants.However,the zeta potential of the nanoparticles was higher than 40 mV,which contributed to the dispersed stability of nanoparticles.In vivo antioxidant study showed that Ast-nano significantly improved the GSH content and the SOD activity in mice model while reduced the MDA and PC levels.The results further proved that Ast-nano was better in protecting against oxidative damage induced by ethanol in mice liver than Ast-macro and Ast-oil.The results indicated that water-dispersible astaxanthin-encapsulated nanosystem was superior to others,which was a high-active antioxidant than astaxanthin oil.
