摘  要:试验旨在优化太行菊总黄酮大孔树脂纯化工艺并研究其降糖活性。采用大孔树脂吸附法,以上样液浓度、上样液速率和洗脱剂流速为自变量,太行菊总黄酮类的回收率为响应值,结合响应面试验对太行菊总黄酮纯化条件进行优化。四氧嘧啶尾静脉注射制备Ⅱ型糖尿病模型小鼠,将糖尿病小鼠随机分成模型组、阳性对照组(盐酸二甲双胍)、太行菊黄酮高剂量组(200 mg/kg)、太行菊黄酮中剂量组(150 mg/kg)和太行菊黄酮低剂量组(100 mg/kg),给药后观察太行菊总黄酮对小鼠空腹血糖水平的影响。结果显示:AB-8型大孔吸附树脂对总黄酮吸附率为37.47%,解吸率达到59.80%;响应面试验表明最佳纯化条件为:上样液浓度2.90 mg/mL、上样液流速1.60 mL/min、乙醇流速1.40 mL/min,此条件下太行菊总黄酮回收率可达81.40%。参比STZ糖尿病模型组,太行菊黄酮高、中、低剂量组使小鼠空腹血糖持续下降并接近正常组。AB-8型树脂对太行菊总黄酮有较好的吸附和洗脱效果,经响应面法优化得到的纯化工艺简单操作性强,且太行菊总黄酮具有明显降低糖尿病小鼠血糖的作用。The purification process of total flavonoids from Opisthopappus taihangensis(FOT) was optimized by macroporous resin, and its hypoglycemic activity was studied. Macroporous resin adsorption was adopted in the experiment. The sample concentration, sample loading rate and eluent flow rate were independent variables, and the recovery rate of FOT was the response value. The purification conditions of FOT were optimized by the response surface test. TypeⅡ diabetic mice model was prepared by tail intravenous injection of alloxan. Diabetic mice were randomly divided into model group, metformin group and FOT of low, medium and high dose groups. The effects of FOT on fasting glucose level in mice were observed after administration. The results showed that the adsorption rate of ab-8 macroporous resin on total flavonoids was 37.47%, and the desorption rate was 59.80%. Response surface analysis showed that the optimal purification conditions were as follows: concentration of sample solution of 2.90 mg/mL, flow rate of sample solution of 1.60 mL/min, and flow rate of ethanol of 1.40 mL/min, under which the total flavonoid recovery rate of FOT was up to 81.40%. Compared with the STZ diabetes model group, the high, medium and low dose groups of FOT continuously reduced the fasting blood glucose of mice and approached the normal group. Ab-8 resin has better adsorption and elution effect on FOT, and the purification process of FOT obtained by response surface method was not only simple but also operable. It was proved that FOT could obviously reduce the blood glucose of diabetic mice.
