摘 要:几丁质是自然界储存量巨大的一种可再生生物资源,降解并脱乙酰后产物为壳聚糖。筛选能够发酵壳聚糖产生酒精的菌株,经过多次筛选得到一株菌K-6,经ITS序列比对、形态以及发酵特性鉴定,确定其应属于酿酒酵母Saccharomyces cerevisiae。实验证明:其能够发酵壳聚糖直接生成酒精,对其发酵条件进行初步优化,在体积分数1%的接种量、壳聚糖质量分数10%的初始条件下,生成酒精的最优发酵条件为温度30℃,pH 6.0,转速150 r/min,时间6 d,其发酵液酒精质量浓度可达到29.1 mg/mL,该研究方向具有较高的研究价值和应用前景,为酒精的生产和几丁质的综合利用找到一条新的可行途径。Chitin is a renewable biological resource with huge storage in nature.Its product after degradation and deacetylation is chitosan.The purpose of this study is to screen a strain that can utilize chitosan to produce alcohol.After repeated screening,a strain of K-6 was obtained.It was identified as Saccharomyces cerevisiae based on the ITS sequence comparison,morphology and fermentation characteristics.It can utilize chitosan to produce alcohol,and the fermentation conditions are optimized.Under the fixed conditions of 1%inoculation amount and 10%chitosan concentration,the optimal fermentation conditions for producing alcohol are 30℃,pH 6.0,150 r/min and 6 days.The ethanol concentration of the fermentation reached 29.1 mg/mL.This work has high research value and application prospect,and it is a new and feasible way for the production of alcohol and the utilization of chitin.
