摘  要:管花肉苁蓉中毛蕊花糖苷含量偏低,利用耐热型β-葡萄糖苷酶可以酶解管花肉苁蓉中的松果菊苷转化为毛蕊花糖苷,以提高毛蕊花糖苷的含量。考察温度、pH、加酶量、酶解时间对松果菊苷酶解率的影响,最后直接对管花肉苁蓉提取物进行酶解,考察反应结果。结果显示:经HPLC测得松果菊苷的酶解率为95.6%;酶解最适反应条件:每克松果菊苷加入0.2 mL的酶液,温度85℃,pH 5.5,反应时间4 h;利用该酶能够将管花肉苁蓉提取物中毛蕊花糖苷含量从1.9%提高至32.1%,进一步利用大孔树脂富集,得到含量64.3%的毛蕊花糖苷粗品。利用该酶能够转化管花肉苁蓉提取物中的松果菊苷变为毛蕊花糖苷,反应效率高,为毛蕊花糖苷的工业化制备提供了一种途径。The content of verbascoside is very low in Cistanche tubulosa,and a kind of thermostableβ-glucosidase can be used to enzymatically hydrolyze echinacoside into verbascoside to improve the content of verbascoside.The effects of temperature,p H,enzyme concentration and reaction time to enzymatic hydrolyzation ratio were inverstigated,then Cistanche tubulosa extracts were directly enzymatically hydrolyzed and the reaction results were investigated.The results show that the enzymatic hydrolysis rate of echinacoside is 95.6%,determined by HPLC.The optimum reaction conditions are as follows:adding 0.2 mL of enzyme solution per gram of echinacoside at 85℃,p H 5.5 and reaction time 4 h.The enzyme can be used to increase the content of verbascoside from 1.9%to 32.1%in Cistanche tubulosa extracts,then enrich verbascoside with macroreticular resin.The content of crude verbascoside is 64.3%.This enzyme can hydrolyze echinacoside from Cistanche tubulosa extracts into verbascoside with high reaction efficiency.The method develops a potential approach for industrial production of verbascoside from Cistanche tubulosa.
