菠萝皮渣中醋酸菌的分离与筛选

摘  要:以菠萝皮渣为样品,采用碳酸钙平板培养基分离,通过观察透明圈,革兰氏染色与产醋酸定性试验,筛选出2株优良醋酸菌。对筛选得到的目的菌株进行产酸定性试验、细菌形态学观察、生理生化鉴定,结果表明为醋酸杆菌属,分别标记为B3和B4。在32℃下以转速150 r/min恒温振荡条件下培养并测定B3和B4产酸量和生长量,结果表明B3,B4在发酵84 h时产醋酸量最高,分别为1.12 g/L(p<0.05)和1.2 g/L(p<0.01)。对其生长量进行比色法测定,表明B3在培养16 h(p<0.05)后进入生长稳定期,B4在培养19 h(p<0.01)后进入稳定期。Isolated acetic acid bacteria with CaCO3-plate in pineapple bran,comparing of the transparent circle,Granmstain and acid qualitative tests to screening 2 strains which had high acetic acied output.It was proved that the strains were acetic acid by the methods of bacterial morphological observation and biochemistry analysis.The propionic acid output and growth of screened strains marked B3 and B4 were cultured in 32℃150 r/min were up to 1.12 g/L and 1.2 g/L respectively after culture during of 84 h.The result of biomass determination indicated that B3 and B4 were up to stable growth period when the culture during was 16 h and 19 h respectively.
