摘  要:以水解圈直径与菌落直径之比(K值)为初筛指标、蛋白酶活力为复筛指标,从菜籽粕中筛选出产高温蛋白酶菌株。RM-2经16S rRNA鉴定后与嗜热脂肪地芽孢杆菌(Geobacillus stearothemophilus)有99%的相似度。当RM-2固态发酵菜籽粕,发酵温度为55℃、料液比为1∶1(g/mL)、发酵时间为24 h时,基质中的多肽得率最高,且显著高于其他较低温度下发酵的多肽得率。The ratio of hydrolysis circle diameter to colony diameter(K value)was used as the primary screening index and the protease activity was used as the re-screening index to screen thermophilic protease strains from rapeseed meal.The similarity between strain RM-2 and Geobacillus stearmophilus was 99% after 16S rRNA identification.When RM-2 was used to ferment rapeseed meal in solid state,the fermentation temperature 55℃,the ratio of material to liquid 1∶1(g/mL)and the fermentation time 24 h,the highest yield of polypeptide was 9.67% which was significantly higher than that of other low temperature fermentation.
